Peptide Method Validation: ICH Q2(R2) and Q14
Research Use Only (RUO). Products and information are intended exclusively for qualified laboratory research. Not for human consumption, clinical treatment or diagnostic use.

A validated method is fit for a stated analytical purpose, within a defined range and sample matrix. ICH Q2(R2) describes how performance is demonstrated; ICH Q14 connects that validation to method development and lifecycle knowledge. A validation label without protocol, criteria, results and method version is not enough for supplier qualification.
Start with the decision the method must support
Method validation begins with intended purpose. Identity confirmation, related-substance control, quantitative assay and residual-solvent testing do not share the same performance requirements. ICH Q2(R2), adopted in its final form in November 2023, frames validation around suitable performance characteristics and a reportable range. ICH Q14 addresses the development knowledge behind the procedure. For a peptide buyer, the useful first question is precise: which specification decision will this result support for this material and lot?
Translate the intended purpose into performance criteria
| Characteristic | Procurement question |
|---|---|
| Specificity or selectivity | Can the procedure distinguish the analyte from expected impurities, degradants and matrix components? |
| Accuracy | How closely does the result agree with an accepted reference across the working range? |
| Precision | How much variation appears across repeat measurements, days, analysts or instruments? |
| Range and response | Is performance demonstrated around the specification or reporting limits? |
| Robustness | Do deliberate small method changes alter the decision? |
| System suitability | What checks show the analytical system is performing before sample results are accepted? |
The required combination depends on the use of the procedure. Avoid demanding every validation characteristic as a generic certificate. Instead, connect each characteristic to risk. A method used only for identity has a different job from a stability-indicating related-substances method. Q2(R2) also allows justified use of prior knowledge and development data, so the evidence package should explain what was tested, what was leveraged and why.
Use Q14 to inspect development knowledge
Q14 introduces an analytical target profile as a concise description of what the method must measure and the performance it must achieve. Development work should identify important method parameters and show how they affect results. For reversed-phase peptide HPLC, examples can include column chemistry, gradient, temperature, mobile-phase composition, wavelength and sample solvent. A wholesale buyer does not need a complete regulatory dossier for every order, but should ask enough to understand method scope, version control and material-specific suitability.
System suitability is a run-level gate
Validation establishes capability under studied conditions. System suitability checks whether the system is ready during a specific run. Relevant criteria may address repeatability, resolution, retention behavior, tailing or signal response, depending on the procedure. The report should show that the defined checks passed before the lot result was accepted. A screenshot of a chromatogram without run context does not answer this question.
Method transfer can expose hidden assumptions
When a procedure moves between laboratories, instruments or columns, Q2(R2) calls for a science- and risk-based evaluation that may involve comparative testing, partial revalidation or full revalidation. Confirm the receiving laboratory, method version, reference materials, sample preparation and acceptance rules. If a supplier and third-party laboratory report different values, investigate the method and sample chain before treating either result as automatically superior. Our third-party testing guide explains custody and report matching.
Request a compact evidence package
- Intended purpose, analyte, sample matrix and reportable range.
- Controlled method identifier, version and change history.
- Validation protocol, predefined criteria, results and conclusion.
- Reference-standard identity and qualification appropriate to the test.
- Run-level system-suitability results tied to the batch analysis.
- Transfer or comparability rationale when the testing laboratory changes.
Keep this request proportional to the material and decision. Review it alongside the quality-control framework and documentation controls. The GHK-Cu 50mg research listing is one example where material-specific identity and assay questions should be stated explicitly rather than inherited from a generic peptide method.

Frequently asked questions
Is an HPLC method validated for every peptide automatically?
No. Suitability depends on the intended purpose, analyte, matrix, range and performance evidence. A platform method may reduce work only when its use is scientifically justified.
What is an analytical target profile?
It is a prospective statement of what an analytical procedure must measure and the performance needed for its intended purpose.
Is system suitability the same as validation?
No. Validation demonstrates method capability. System suitability is a run-level check that the analytical system is performing acceptably when samples are tested.
What should be checked after method transfer?
Check the receiving laboratory, method version, instrument and column equivalence, reference materials, comparative results, predefined criteria and revalidation rationale.
